It sounds like dude had an issue with the employee from the start just because they presented themselves as the opposite sex.

I would have probably just stared at him until he started to bag up my purchases, and hand him my cash/card while not saying a single word, and not breaking eye contact until I get my receipt.
Kind of like a stone face, irl.

I said no thanks he replied with the smart comment then started taking his time to ring me out like it was a problem I didn't donate $6 and that's when I spoke up as he was saying stuff under his breath as other people behind me are getting annoyed its taking this dude all this time to check out a button down shirt for $26.
If that makes me pretty, soft, emotional, catty, etc then okay lol

Pretty much. You felt embarrassed because the dude made you look broke or cheap or not a very giving person? Is that it? Had there been no one else in line would you have reacted the same? Maybe you were having a bad day or something but every cool and chill person I know would not have reacted that way.
